You take it about 45 minutes ahead; most people feel it within the hour, and the effect can linger for several hours. The one thing to expect is nausea - it's the most common side effect, worst on your first try and at higher doses, and it usually settles as you get used to it. How it actually works
Bremelanotide - the deaminated metabolite of melanotan II, and MC4R-selective where its parent is not. It acts centrally, in the hypothalamus, on the pathways that generate sexual arousal.
For stacking, two things follow and both are commonly misunderstood. It is not a vascular drug: PDE5 inhibitors act peripherally on blood-vessel smooth muscle, this acts on the brain, and they are genuinely different mechanisms rather than two versions of the same one. And because it is a melanotan II metabolite, running both is hitting MC4R twice.
What people report, in full
PT-141's feedback splits along the same line as its approval status. For women (where it's FDA-approved), roughly 40–50% report a real boost in arousal; for men (off-label), a bit more - around 50–60% - report better erections and desire. It kicks in within 30–60 minutes and lasts several hours. The one side effect to plan for is nausea, which hits about a fifth to a quarter of users and is often headed off with an antihistamine beforehand. It's an as-needed thing, not a daily one.
