How it actually works
A sixteen-amino-acid peptide encoded not in the cell nucleus but in mitochondrial DNA, within the 12S rRNA region - one of a small family of mitochondrial-derived peptides. It activates AMPK, the cell's low-energy sensor, which shifts metabolism toward fatty-acid oxidation and glucose uptake and away from storage.
For stacking, AMPK is a well-populated crossroads: it is the same node metformin acts on, and exercise and fasting activate it too. That does not make combining them dangerous, but it does mean the effects are unlikely to be simply additive. SS-31 is the more complementary pairing, because it works on mitochondrial structure rather than on the same signalling switch.

The dosing-unit argument that ran through the first half of the year has largely settled, and it settled on milligrams. Real practice now clusters between about 1mg and 5mg per shot, and 0.25mg is the amount people describe feeling nothing on. Worth knowing that our 5mg three times a week sits at the top of that range rather than in the middle - plenty of people run 1-2.5mg and report the same thing.
